All you have to do is to use OpenIV which will create a mod folder for you. This, will let you play modded game files, which will not be sourced when running the Online version of the game.

Not sure if this is helpful, but outdated graphics mods can also prevent GTA from launching. I once forgot to update VisualV after the .exe was replaced and I was being kicked out of the game after, like, two seconds.
